Capture and Detection in a Single Step
In conventional sandwich ELISA, the capture antibody is pre-coated on the well and binds to a selected antigen present in a sample (hence the’sandwich’). After wash steps, the detector antibody binds to a second site on the antigen to form a complex. The plate is then incubated with a substrate that converts the antibody-antigen complex into a color, fluorescence or electrochemical signal. The plate is washed and the signal is measured to determine the concentration of the analyte.
